Characteristics of Bone-Like Apatite Containing Bovine Serum Albumin Coated on Titanium

Abstract
In this study, the characteristics of the bovine serum albumin (BSA)-containing bone-like apatite (BLAp) coated on titanium by immersion in simulated body fluid (SBF) were investigated. Titanium disks used as substrates were treated using anodic oxidation and hydrothermal treatments. A mSBF (modified-SBF) which contains twice of the concentration of Ca and P ions compared with that of standard SBF was used for coating BLAp layer. BSAs were premixed with a CaCl2 and a KH2PO4 solutions, respectively, in order to prepare Ca-mSBF and P-mSBF. The S-BLAp, Ca-BLAp, and P-BLAp groups were fabricated by soaking titanium specimens in mSBF, Ca-mSBF, and P-mSBF, respectively. The morphology of the crystallites observed in the Ca-BLAp and P-BLAp groups was more rounded compared with that of the S-BLAp group. In the cross-sectional images of BLAp layers, the S-BLAp and Ca-BLAp groups showed the well-aligned crystallites, while the P-BLAp group showed the randomly aligned crystallites. The Hydroxyapatite (HAp) crystalline structure was dominant for all groups. The XRD patterns of the Ca-BLAp group showed that HAp crystals were aligned along the c-axis. The crystallinity of P-BLAp group was lower than that of the Ca-BLAp group. The amide peaks related to BSA were identified on FT-IR spectra for the BSA-containing BLAp groups. The amount of dissolution of Ca and P ions was high in the S-BLAp group, while that was low in the Ca-BLAp group. After the dissolution experiment, amide peaks were still observed in the Ca-BLAp and P-BLAp groups, which inferred that BSAs were released slowly in BSA-containing BLAp groups.
